Imatinib and mechanism of action of Resveratol on IM resistant cell lines

Categories:

Hi Sandy,

There was mention in the press recently of the potential benefits of dosing with Reservatol for patients in the early stages of dementia of the  Alzheimer type.

Some years ago there was the beginnings of a thread on this forum asking for details of studies that related to the therapeutic use of Reservatol in cases of imatinib resistance.

Resveratol is a food supplement derived from Japanese Knotweed (polygonum cuspidatum) and in addition red grapeskins; the benefits of polyphenols is normally associated with improved cardiovascular performance  and provides the body with strong anti-oxidants.Other sources are grapeseed extract and bilberry.

I have found 3 articles which explore the benefits of Reservatol on IM resistant cells:

1.Imatinib mesylate -resistant human chronic myelogenous leukemia cell lines exhibit high sensitivity to the phytoalexin resveratol -Puissant et al ,The Faseb Journal v.22, 2008

2.Resveratol promotes autophagic cell death in Chronic Myelogenous Leukemia cells via JNK- mediated p62/SQSTM1expression and AMPKactivation- Puissant et al ;Cancer Research, Jan. 2010

3.Apoptotic effects of Reservatol,a grape polyphenol on imatinib-sensitive and resistant K562chronic myeloid leukemia cells-Can et al ; Anticancer Research v.32 ,2012

My own observations are that these seem to be lab based studies and to date no further evidence exists on the benefits of Resveratol for CML patients ?

In addition would the potential benefits of this food supplement be more relevant to drug resistance developing after long exposure to imatinib or to potential IM failure in the early stages of treatment?

The usual queries apply on the safe daily dose for this food supplement and the potential  hepatatic and renal side effects that may arise.

Does anyone have any further evidence?

With best wishes

John
